Effects of Cycloheximide on Glutamic-Oxaloacetic Transaminase Isozyme in Different Organs of the Mouse


Abstract
Effects of cycloheximide on glutamic-oxaloacetic transaminase(GOT) isozyme in different organs (e.g., liver, heart and kidney) of the albino mouse have been investigated. The activities were determined on the whole homogenates and sub cellular fractions separated by differential centrifugation after the intraperitoneal injection of cycloheximide (1mg/kg) into mice.
The activity of glutamic-oxaloacetic transaminase isozyme was assayed by colorimetric method of Reitiuan-Frankel.
1. Ninety percent of the glutamic-oxaloacetic transaminase isozyme in different organs was found to be localized in the supernatant fraction with the separation of differential centrifugation as described in the text. And it was found that 10% of glutamic-oxaloacetic transaminase isozyme exists in the mitochondrial fraction.
2. The supernatant (extramitochondrial) isohyets was inhibited 20-30% at 1. 5 to 4 hours after the intraperitoneal injection of cycloheximide (1mg/kg) into mice. By contrast, the mitochondrial one was not affected at all by cycloheximide.
From these results, it was suggested that the diminution of the supernatant isozyme activity might be caused by the inhibition of the biosynthesis of the enzyme. GOT. And. the supernatant isozyme of the different organs (e.g., liver, heart and kidney) of the albino mouse is more regulatory than the mitochondrial one.
